I host a blog on my Dreamhost account. It started out as a subsection of a website and it’s address is mydomain.com/blog. I’d like to give it its own domain but I don’t want to physically move it out of its current directory structure. Is it possible to register a new domain for (ie. blog.com) it and have that work as well as the old address (mydomain.com/blog)?

Yes, you can change the home directory of your domain via DH panel --> Domains --> Manage Domains --> Web Hosting --> Edit
Change home directory from /home/username/domain.com to /home/username/domain.com/blog

A subdomain might be logical, like blog.yourdomain.com, rather than registering a separate domain.
